The Medite USE 33 Ultrasonic Decalcifying Bath uses ultrasonic power and temperature control to facilitate significant reductions in the time required to prepare and decalcify bone and tissue samples.

The ultrasonice decalcifying bath promotes the rapid destruction of crystalline structures like calcum phosphate, magnesium phosphate and calcium carbonate. In combination with appropriate solutions it provides maximum cell tissue preservation and a significantly accelerated diffusion process..

The USE 33 maintains the tissue samples at 17 degrees C during the process which prevents the samples from being warmed excessively by the chemical reaction of the decalcifying solution. This guarantees 100% preservation of the morphological structures and the antigenity of the samples. No artefacts are generated from shrinking or swelling and all further histological and immuno-histochemical methods can be applied to the samples

The decalcifying procedure can be carried out with different solutions. A quick result can be obtained with strong mineral acids (such as hydrochloric, nitric or formic acid). Their use, however, may result in the loss of morphologic and/or immuno-histochemical information. With the USE 33 these acids work successfully within 15-60 minutes in numerous kinds of specimens.

An alternative to the mineral acids is available in the form of a special calcium ion binding solution (USEDCALC). These solutions provide optimum conservaton of all tissue structures under much milder conditions.

Decalcifying times can be reduced by 60 - 65 percent compared with conventional procedures. The ultrasonic burst opens the crystal structures containing calcium resulting in faster removal and binding of Ca2+ ions in the outer solution. The strong integrated cooling protects the tissue from temperature rises caused by the chemical reaction

High performance fixation of the specimens can be made in the instrument. The ultrasound accelerates the diffusion speed of the fixation molecules into the target tissue. This is dependent on the molecular structure. For example glutardialdahyde invades much slower into the tissue than formaldehyde. With the USE 33 a significantly faster fixation is achieved compared to the conventional method. A great number of bone containing specimens can be fixed in an overnight process with safe and reliable success.

The USE 33 also supports clearing and cleaning procedures integrated into the process. Surplus fixation solution in the tissue will be rapidly transferred into the clearing solution. Even primary dehydraton procedure time will be optimised.

Fixation in 10 vol.% formaldehyde - 3 to 6 hours

Fixation in 10 vol.% formaldehyde - 2 to 4 hours
Clearing in running water - 15 min
Decalcifying medite new decalc - 15 to 60 mins
Cell protection - Na2SO4/Li2SO4 - 3 to 6 hours (optional)
Clearing in running water - 15 to 30 mins

This represents, at the lower end, a saving of 10 hours 45 minustes (or 67%) and, at the higher end, a saving of 22 hours and 45 minutes (or 64%)
